概要 | Vacuolar type ATPase (V-ATPase) is a primary proton pump with versatile function in various tissues. In nerve cells, V-ATPase is required for accumulation of neurotransmitters into secretory vesicles and subsequent release at synapse. Neurons express a specific isoform (G2) for the G subunit of V-ATPase constituting the catalytic sector of enzyme complex. We generated a mouse lacking the functional G2 by gene targeting. G2 null animals exhibited none of apparent disorders in their architecture and behavior. In brain of G2-null mouse, G1 subunit isoform, which is ubiquitously expressed in neuronal and non-neuronal tissues, is more abundantly accumulated comparing to the wild-type animals. This upregulation of the G1 was not accompanied with the increase of mRNA. These results indicated that the function of neuron specific G2 isoform is compensated by an increase of the G1 isoform, whose accumulation levels were controlled by post-transcriptional regulatory mechanism. |
